I just bought a 1978 Parker Bros edition that is missing one of the tiles.
I have: 21 red 10 yellow 16 orange
I assume that I am missing a red tile. Could someone please confirm this?

weird. i have 24 red, 13 orange and 11 yellow. i verified that my sets instructions stated 48 markers, no "per color" count.

funny thing. this morning, i found a copy at a yard sale with sealed bits. if i can move them around within the baggies, i'll give you a good piece count.

welp, just to further baffle you, my sealed bags contain the following...
18 orange 13 yellow 23 red
54 total, 6 over the piece count of 48.
there are folks offering parts to complete your set, whatever "complete" means in this game

My set (bought new) has the following:
18 orange (2 each of 9 different patterns) 11 yellow (10 and 1 extra) 21 red (20 and 1 extra) -- 48 and 2 extra (according to publicity information at the time)
In play I have never seen anyone use more than 7 orange pairs and probably no more than 8 yellow or 16 red
Contents should also include 2 part plastic black box 1 cardboard outer jacket (slides over box) rule book wipe-off plastic coated cardboard solution board 1 black crayon several yellow balls (At least 4, but I think 5)
It sounds like you are missing two orange tiles. The red tiles indicate a hit and so are used individually. The yellow indicate a reflection, a place where a ray goes in and returns out at the same point and so are also used singly. The orange are used for a ray that goes in at one location and come out at another, and thus are used in pairs. If you are missing two, then you should have at least 7 usable pairs - which should be enough to play. If you have 8 usable pairs, and one entire pair missing - the following might help you figure out what you have.
Square - Circle - Triangle - Hourglass - Starburst X (with circular ends) - Plus (not larger at ends) Diagonals (three, middle longer)- Dual Curves (outline circle)
The first five are open shapes; the other four are more like drawn fat lines. If you have the outer jacket, the large picture on the back shows all of these but the first three (Square, Circle, Triangle) though someone seems to have knocked off one of the Plus signs. All of them are pictured in the small picture in the lower right - but you have to look close to see the Circle at the very middle of the back.
I hope that helps.

further adding to the confusion:
13 Yellow Squares 29 Red Squares 21 Orange pieces(9 pairs and 3 duplicates - or is that 6 pairs and 3 triplets?!) ____ 63 total Plus 5 yellow balls, 1 black crayon, & 1 eraseable scorecard, in my newly acquired thrift copy

So, the consensus seems to be something like this:
Black Box - Parker Brothers 1978 edition contents:
