If I have a white meeple and put it on a promotion space on my player board can I turn that white meeple into the color meeple I need for my milestone card? Or do I have to have that color meeple already available?

You need to have it available..the white worker only does the promotion.

You need to put that worker on the milestone card. Remeber that those workers are used up and have to be put back into the reserve.

But remember you can't have less then 4 workers so if you have to remove one because of fullfilling a milestone you have at least 5 workers

I confirm you need to have the meeple available.

For reference, rulebook page 11:
Quote:
Promoting a Worker means to move it from your supply or your Player Board and place it on one of your company Milestones.


The only way to turn a meeple into another color is with the "Exchange a worker" bonus (the one with 2 workers and an arrow). For example, using the Purple special action when building a telegraph office.
