Given that an open door action interrupts movement, and an interrupt requires a large monster to expand...

does a large monster need to have available space to expand to be able to open a door?

For example, in the picture if the red X has a figure on it, can a 2X2 monster open that door?

Correct. in order to interrupt a move actioj with another action, a large monster must expand. If it cannot expand, it cannot interrupt. It is one of the few downsides to large monster movement (and can sometimes be an advantage).
