In the MegaMania rulebook, it mentions that there are optional, untested rules for playing the game with more than four players. Has anyone tried it? Any advice?

The only issue would be the colour of the additional block counter. BlockMania+MegaMania gives you 4 blocks/4colours, but you'd probably need to make a set of alternate coloured counters to add more block boards...I don't think that would be a big deal, though.

The gameplay would just be more chaotic...but while the skyrails and megway are intact, people can zip between buildings with ease no matter how many blocks...and you could make a rule that the left-most and right-most blocks "wrap-around" as though the blocks are in a circle.
