My used copy is missing red men #3 and #5. Is it still playable?

Should be. You'll just be down two subs. If either of these was a position player i.e. Blitzer, Thrower etc. just paint the coloured band onto a free Lineman. Use any other two miniature to stand in for the missing men if you need.
