We really like Hansa Teutonica and the Eastern Expansion. We have played it with all different numbers, but personally I like it most with 4+. As we are often 6 persons, I am thinking of playing it with 6 persons. Has ever someone tried it this way? I know I need a 6th personal player board, but other than this, do you think of other complications with 6 players which hamper the playability?

You can certainly try it, but I am not sure it would make a good game. Optimal play by all parties seems like it would result in gridlock on all of the skill tracks, and possibly all of the bonus marker tracks.
