I'm confused about the seperate, 'at game start' & ,'Draw Stack' piles

I am clear about at the very begenning of the game where the, 'at game start' tiles come into play & by themselves but then do I:

2 player game example: during the remaining four phases do I shuffle ALL 1,2,4,6,8,10 throughout the game? are the tiles 2,10 only used at the very beginning?

Yes. You will use ALL tiles over the rest of the game. After the first auction, the tiles 2 and 10 will be shuffled with the draw stack.
If the tiles bought were say, 6 and 8, then the draw stack will next be comprised of tiles 1,2,4 and 10 for the second round.
