AH accomodates drop-ins quite well, and Sister Mary even moreso:

Sister Mary is my favorite...
but not of her ability but her story and the feeling of playing a holy character against the horrors of Lovecraft.
We always pick characters based on the story they have and not based on what they do.
P.S. I really would like to know how many in here actually remember (if ever read) the story at the back of the character sheet.

Darrell! The retainer is great, but I love the extra incentive to have location encounters.

@ MountainRoot, I really enjoy Sister Mary's story, too. I think she was actually the first investigator I ever played, and it was because of her story. I've started to just play randomly-drawn characters, but how much I like playing them is usually a direct correlation to how much their background "speaks to me."
My absolute favorite character is Ursula Downs, from Innsmouth. I am a sucker for archaeologists/anthropologists, and she's just got so many great features: Starting anywhere she wants, 4 clue tokens right off the bat, getting to use location abilities and STILL have an encounter that turn, spending one measly clue token to cancel an encounter. She is badassery personified. Playing with her personal story cards is also fun, because there is an element of luck with her pass/fail conditions that not all other investigators have, which keeps things interesting.

One of the best games I had was an almost lost game that Wendy, single-handedly, turned into a glorious victory. She ran out from the bank, sneaked past a heap of deadly monsters, and saved the day - while the big guys went in and out of hospitals and asylums.

I've got 3: Joe Diamond, Carolyn Fern, and Hank Samson. Joe's got just the right combination of starting gear, stats, and great special ability to be a good all-rounder. Carolyn's hands down one of the best spellcasters in the game. And Hank's just a lovable meatstick.

I don't have a lot of experience with this game. I have only played a fraction of the characters available. Since we have started to play this with only four investigators, instead of trying to get six or seven people together, I hope to get some more games in and to play even more characters.
Of the limited number of characters I have played. I have had a lot of fun playing Wilson Richards. He starts with a motorcycle and a decent weapon. Which makes it easy for him to travel across the board and clear the streets, so that other investigators can get where they need to go. His ability to gain a dollar instead of encountering the space has been very helpful, and he has infinite focus.
