The title says it all:
Seeking a cooperative game, highly replayable, hard to win ...

(Hard to win in my case: AH and Legends of Andor for me are very easy to win)

I would say Robinson Crusoe: Adventures on the Cursed Island definitely fits your description.

Though I have only played it once, that's the first title that came to my mind.

In addition to the excellent suggestions above, I would also recommend
The Lord of the Rings: the card game.

And if you want it REALLY hard, get the expansion right away too.....

A Touch of Evil has SEVERAL difficultly level modes. If you want hard, you definitely will have it on this one. The hardest is VERY TOUGH.

Pandemic. Haha, just kidding

If you want hard, try Defenders of the Realm: http://boardgamegeek.com/boardgame/65532/defenders-of-the-re...
It is absolutely brutal. In that game each player has a hero jointly charged with defending the center board space which is 'The Town'. There are 4 Monster Lords or some such that are progressing to the center of the board from the outskirts all the while spawning minions on the other spaces of the boards. The Lords move forward based on cards drawn: if the Monster Lord is on the shown Space, it moves forward to the next one. Fighting a Monster Lord is quite the ordeal! You have to succeed on some pretty hard die rolls, though you get extra dice by discarding cards collected thus far on each turn corresponding to the Monster Lord you are fighting (red, blue, etc.)

This game has not been beaten by my friends and I (which might say more about me and my friends than the game haha). Maybe someone can corroborate the difficulty of this game?

I've also found it quite fun to replay, but I'm not sure how inclined to play it my friends and I would be with just us if we already beat it.

Defenders of the Realm

There's varying degrees of difficulty.

Space Hulk: Death Angel – The Card Game

There's a lot of game packed into this small package.

In 5 plays, my friend and I have not even gotten close to winning Ghost Stories. In a game where the goal is to get all the way through the deck of ghosts and beat the big bad guy at the end, I think our best attempt was our most recent one where we got 2/3 of the way through.

I don't think you can beat Ghost Stories for the combination of hard and replayable. Ghost Stories: White Moon increases the variety, but decreases the difficulty a bit.

Witch of Salem is another good choice.

If you add in the expansion and crank up the difficulty options, Space Alert can be pretty brutal, but it is primarily a skill game, so in theory you can eventually get good enough to match the difficulty.

Say Bye to the Villains is downright nasty, and fun, but the replay value seems a lot lower than any of the others I listed above.

I've got a long list of co-ops rated in this geeklist, including my ratings for replayability and difficulty.

Adding my vote for Defenders of the Realm. It's the game I get the most satisfaction out of beating! One of the things I like is that the Richard Launius, the designer, has posted some extra print and play material in case you get to the point where you need to see something new. The base game hasn't gotten old to me yet, but sometimes I play with some of the P&P stuff because they add some new mechanics and some heavier theme to go with it.

We're up to 7 plays and have seen the boss once. Then we died.

Nice list, but lost all credibility when I saw your opinion about the replayability of robinson crusoe just because don't like the placement worker mechanic...
but thx to this list:
Now i see Yggdrasil too
