So in page 23 of the rulebook under DEFEATING CREATURES:

During the end phase, when the number of Damage Cards dealt beside a Creature Card is equal to or greater than the creature's Health value, the creature is defeated.

But the quick start guide under PHASE 4: CHECK FOR VICTORY:

During this phase, check to see if the total number of damage cards next to either creature card equals or exceeds the creature's total [b]HEALTH AND ARMOR. If so that creature is defeated and removed from the play area.

So the question is whether or not damage cards are supposed to equal health or health+armor.

And the tldr version: does it take 4 or 6 damage to kill calamity.

The quickstart rules are a simplified version of the game, intended to help players get a handle on phases and maneuvering, and those rules do not all carry over to the actual game; as you noticed, there are differences. Wherever you see those differences, you should assume that the quickstart is in error.
