I nearly exclusively play the Overlord.
A few of the combinations I consider most effective, as in "the combinations that make my day the hardest" (base game only):

Widow Tarha as Runemaster
Her Reroll Ability and Double Attack Feat sync up nicely with being the strongest ranged Damage Dealer in the box

Jaine Fairwood as Thief
Tumble combined with her heroic feat and high awareness basically makes her unblockable, unstoppable and overall annoying.

Syndrael as Berserker
Giving the group piles of mobility, being very mobile herself and the ability to immobilize at will. Super Annoying for me as the OL

Avric Allbright as Disciple
Just forget about killing anyone if you don't have the monsters and cards to do it during one round with this superpowered healer on the board.

Haven't played it but Elder Mok as bard is apparently a beast because of how much synergy the skills have with his hero ability.

Jaes as the Runemaster is a boss. Especially with inscribe rune, exploding rune, and quick casting. You can give him swords that suddenly do blast attacks!
