Has anyone tried playing this with 5-6 players? Any suggestions on balance? Assuming we find/make other colored tokens to use, at the very least it makes sense for us to would increase the number of cards on the board at once, like you do from 3 to 4 players. Anything else?

While I haven't tried it, I would assume that -- since really the only direct effect that players can have on your game play is drafting a card that you wanted -- increasing the number of cards should suffice to offset the increase in the number of players.

That being said, I asked a similar question during the Kickstarter campaign, and Adrian came back saying that it had not been playtested for 5 or more players, and was not recommended -- as I recall -- due to downtime issues. Having now played the game several times, I can certainly appreciate that, as the downtime can get pretty intense, especially during the beginning (when you have more hourglasses to spend) and end (when each hourglass matters more) of the game.

Even with 4 is quite long, I wouldn't either recommend it with 5 or 6, although that's perfectly doable. I feel that the sweet spot is 2-3 players. 2-player games can be really fast and 3-player games are quite good, too. Having said that, I'd also play it with 4 players as well, contrary to other games that I really love but are not for such player counts.
