Cruise missiles capability for each side is given in scenario setup and reinforcement tables.

How should cruise missile platforms such as B-52s or (in the new supplement) US and PRC subs be considered? Do they grant another cruise missile factor each? Only once or for each turn?

As specified in the Game Specific Rules, each platform provides the ability to use a Cruise Missile Point at particular times during the Sequence of Play (see 15.0 in the NWT GSR).

Submarine Markers from the supplement are treated differently as specified in 1.2.3 of the Supplement Submarine Marker rules. They do not use CM Points.
