Hey guys, just wondering my bird is perched and I set him in the 'hey I'm going to take off' position, can I still change his tilt?

If not... can I do a piloting check and then tilt?

Your bird has to start at center tilt, but you can do the "beginning of movement piloting check" to change his (or her) tilt as you surmised.

The ball-and-socket joint is not designed to tilt left or right while the bird is in the launch position. If you try it, you'll likely break the female part of the joint.
