Bonus = Prepare a room of your choice without paying any cost (all other room preparation rules apply).
Penalty = Remove an unoccupied Room Tile from the highest possible row of your hotel, or lose 5 VP.

For example,
I have an occupied room tile in the first row and an unoccupied room tile in the second row. And, I have an occupied room tile in the third row.

In this case, is it possible to remove an unoccupied room tile in the second tile?

No, doesn't matter. You just have to build off of the connections afterwards.

Can you build adjacent to any room you've already built, or do you have to build where you can trace a path back to the starting room?
