A rules question that has come up a couple of times in my games. All the examples for the transport tiles show having two and getting four points. What I have wondered is whether it is possible to score more than four, for having three or more tiles all separated, as in the following image - does this score six? And would having four tiles in the four corners of the 4x3 square score eight?

Yes, two points for each Metro station...after you have at least two stations. Thematically, that makes sense because you need two stations for the trains to travel between. Having more destinations each scores two points, assuming they're not adjacent.

1 station = 0 points
2 stations = 4 points
3 stations = 6 points
4 stations = 8 points
etc.
